Flashing from manual trans to a 4L60E

I have a V1 that is setup for a manual trans. can I flash it to auto without buying a new tune

Yes.

You can flash in any OS/tune that is compatible with your PCM, and you can do segment swaps.

What year/model/vehicle do you have...?

In V7 scantool or V7 tunetool, go Help->Register EFILive and post a screenshot (blot out the serial number).

When you say post a screenshot and blot out the serial number you mean here on this Forum, and do you mean the serial number for the cable data link. I'm sorry for al the question but I'm just learning. That's why I'm glad that this forum is here.

So everyone does not see your serial number of your V1 (if it matters, maybe it doesn't)...
