I'm working on a Gen V 4.3 with an E92 controller, and a 6L80E w/T43 Tecm. Is there a good chance I can make these work together? If not, what challenges might I face? The 6L80E is from an '11 Camaro SS, the engine from a '14 Silverado and I do have an Autocal and software. Thanks for any help.

Your options will be to find a T43 match, there isn't much you can do about the E92 as I think 2014 was the first year it came out, the 8 speed didn't hit until the 2015 models.
So you have a narrow window to get a T43 OS that will match to the ECM, I guess the 2014 4.3 might have come with a 6 speed Auto anyway?

The engine and ECM are from the same 2014 truck, the 6L80E is from a 2010 Camaro SS. Fortunately the truck was also 6L80E equipped, so that may be my saving grace. I have one more quick question, I wonder if it's possible to make use of the boost parameters that are available in the E92?

You will need to match the TCM OS to what was in the truck (well, to what the ECM expects), the CAN messages the ECM and TCM transmit to each other all the time are not the same for every year model or model vehicle, you will have issues unless you get them matched.
I'm not sure how many deep down parameters and tables are in the ECM to enable an NA tune to be able to control turbo wastegates or supercharging, probably a very deep rabbit hole
