I've tuned 3 of them, so you should be able to.

Check what OS it is running by entering the VIN at http://tis2web.service.gm.com/tis2web

Then search your Calibrations folder for that OS. Make sure you have the most up to date version of EFI Live installed.

I have also done a few 06 silverado's all had 5.3's.
