Ok, so I found a '02 Yukon P01 tune with Flex Fuel active (OS 12216125). I think the steps are:
1. Full flash COS3
2. Flash calibrations-only from the Yukon tune.
3. Read the tune
4. Run script to program COS3/Yukon tune with most current calibration
Is this correct?

You lose me at #4. Once the tune is read follow the tutorial to make sure all the other values are set correctly and then perform another calibration flash. But there's a bigger problem here: that OS doesn't support a COS as far as I can tell. If it did, I'd have been on it eons ago. There is all the CAX stuff to flip the flex fuel switch on a P59 though and that does have an OS that is supported by COS3. I added the flex fuel stuff to my Z06 which has a P59. Still have yet to buy a sensor though.

You're right about OS 12216125, I forgot. So, I remember seeing a Sloppy Mechanics PCM video listing the capabilities of the P01 and P59. I thought you could get 2bar boost and flex fuel capability from the P01.
Maybe that's only with HP Tuners?

Just trying to figure out if I can get boost and Flex fueling from a P01 PCM...
