I started with 12212156 and went to COS 2020001. I am now swapping from a 4l60e to 4l80e do I have to go back and make a new tune with the 12212156 and then change to the custom operating system again?

You can use the 4l80 OS and do the 2020001 COS then do a Segment swap for the 4l80 into your current tune.
