Default 4l80 swap for custom operating system

I'm doing a 4l80 swap to my 2005 1500 Sierra , can I do a custom operation system after doing the segment swap for my stock tune.
My setup is as follow :
    418 ls1
    Custom intake
    Twinturbo
    3bar map sensor
    #80 injectors
Cusome fuel system with surge tank

I'm planing to run 2 bar of boost.

    Thanks in advance

I imagine the 2500 trucks had a 80E behind them, should be able to find a stock file with an 80E and either do a seg swap or leave it alone then do a COS upgrade.

Yeah, custom os is just a base for your original os that adds some additional parameters .
So build a .tun with seg swap then follow the custom os tutorial and your good to go.

These guys are right do the segment swap first then do the COS.
Don't loose any sleep over it, just take your time and do it right.
    I've got a L31 350, 4L80E, 12200411 PCM with COS5.

Back from the dead! Just to clarify now, once I swap those 2 wires and do this segment swap, the 4L80E should work without any issues, right?

I pulled two wires out of the trans harness (that the 80 doesn't use) and used them for the additional
speed sensor (that the 60 doesn't use) Basically both transmissions have the same number of wires total.
The 60 has one more solenoid than the 80 and the 80 has one more sensor than the 60.
So you have to swap those wires at the PCM and at the trans.

The wiring differences between 4L60/65/70E and 4L80/85E can be summarized as follows:

4L60/65/70E have two additional solenoids: 3-2 solenoid and TCC Clutch/Enable solenoid (wired via transmission internal harness).

4L80/85E have an additional speed sensor: ISS (wired via external harness).

Some 4L65/70E also have the ISS (wired via external harness).

The TCC PWM signal changes pins (wired via transmission internal harness).

PNP switch: some PCM's use 1-wire PNP while others use 4-wire PNP (all cases wired externally).
